Toxicosis por cobre (ATP7A, ATP7B y RETN)

The mutations analysed in this test are the mutation for Wilson Disease (ATP7B-related) as well as the two modifiers of copper toxicosis (ATP7A- and RETN-related). Information about individual tests in this package is available in the section ‘Included Tests’ on this page.
Copper levels in the body are regulated by both dietary intake and secretion from specific bodily organs, such as the bile ducts. Multiple genes play crucial roles in maintaining optimal copper levels. A mutation in the ATP7B gene is associated with increased copper accumulation, leading to Wilson Disease. Conversely, mutations in modifier genes ATP7A or RETN appear to confer a protective effect against copper buildup in the liver, formerly known as Menkes Disease. Currently, these modifier genes are only relevant for Labrador Retrievers. However, the ATP7B mutation can cause clinical symptoms in several dog breeds.

MDR1 Multi Resistencia a Medicamentos – Perro

La Multi Resistencia a Fármacos 1 o Sensibilidad Multifármaco parece heredarse de forma autosómica dominante con penetrancia incompleta. Los perros afectados son susceptibles a la neurotoxicidad después de la administración de ciertos medicamentos de uso común como la ivermectina. La afección está relacionada con el gen del miembro 1 de la subfamilia B del casete de unión a ATP (ABCB1), también conocido como gen de resistencia a múltiples fármacos (MDR1). Este gen codifica una proteína transportadora de fármacos llamada P-glicoproteína (P-gp), que desempeña un papel crucial en la barrera hematoencefálica al evitar que ciertos fármacos se acumulen en el cerebro. La variación genética que está presente en un animal, se origina en ambos padres. La mitad de la variación se origina en el padre, mientras que la otra mitad proviene de la madre.

Para la verificación de parentesco, típicamente se visualizan de 20 a 40 características genéticas. En este proceso se mide la longitud de los fragmentos genéticos. La longitud en la medida de una característica genética en un descendiente debe corresponder a la longitud en la madre y el padre que se proporcionaron para comparación. En estos dos ejemplos, se muestra cómo se aplican las reglas básicas en la verificación de parentesco.

En la figura se proporciona un ejemplo de un parentesco correcto. En esta figura, se muestra el ADN de tres individuos: una descendiente (línea superior), una madre potencial (línea media) y un padre potencial (línea inferior). En cada línea se muestra un marcador genético. Dos fragmentos de ADN son visibles como picos. El primer fragmento de la descendencia se origina en el padre (la longitud del fragmento es de 150), mientras que el segundo fragmento proviene de la madre (longitud del fragmento 152). En este caso, ambos fragmentos de la descendencia están presentes en los padres: la paternidad es correcta.

En el segundo ejemplo, se muestra una situación donde la filiación no califica como tal. Las tres líneas se muestran en el orden de descendencia, madre potencial y padre potencial. Nuevamente en cada línea se muestra un marcador de ADN, donde dos fragmentos de ADN son visibles como picos. El segundo fragmento de la descendencia está presente en la madre (longitud del fragmento 152), mientras que el primer fragmento en la descendencia (longitud del fragmento 150) NO está presente en el padre asignado. En este caso, un fragmento está presente en la descendencia, que no está presente en ninguno de los padres: la filiación no califica.

Cuando se comprueban de 20 a 40 fragmentos genéticos diferentes, la probabilidad de que no se detecte un parentesco incorrecto es muy pequeña. Los fragmentos genéticos que se utilizan para la verificación e identificación de parentesco no proporcionan información sobre propiedades como el color y la calidad de un animal, planta o humano, ya que los fragmentos no están codificados.

Cuando se mide la longitud de varios fragmentos de ADN para una muestra, se establece un perfil de ADN. Polidactilia (3 variantes) – Gato

La polidactilia es una afección anatómica en la que un animal tiene más dedos de lo habitual. Se conocen múltiples variantes, la variante más conocida es la “mutación de Hemingway” que se produce en los Main Coons, también conocidos como Hw.

La polidactilia en los gatos, un defecto congénito, resulta en más de 18 dedos. Los dedos adicionales pueden aparecer en cualquier pata, en varias combinaciones, y su aparición sigue un patrón estadístico irregular. La polidactilia no influye en la salud general de un gato. No es doloroso ni dañino y no conduce a más problemas de salud.

En CombiBreed, las tres variantes anteriores se prueban juntas como un paquete. Se darán los resultados de las tres mutaciones. Cariotipo – Caballo

En los equinos, el cariotipo se usa comúnmente para diagnosticar las causas genéticas de la fertilidad reducida. Si hay anomalías cromosómicas, el cariograma proporciona una indicación clara, lo que facilita intervenciones específicas o decisiones informadas sobre la reproducción para mitigar los problemas reproductivos.

El cariotipo permite la visualización de un conjunto completo de cromosomas dentro de una célula utilizando un microscopio óptico. El proceso consiste en aislar células de la sangre del caballo y organizar sus cromosomas en un cariograma estandarizado para un análisis detallado. Al aplicar bandas G (tinción de Giemsa), surge un patrón de bandas distintivo, lo que permite la identificación de anomalías cromosómicas. Estas anomalías pueden incluir trisomías, deleciones, inserciones, hermafroditismo y translocaciones recíprocas y robertsonianas.
